What is Urolithins B?
Urolithin B( CAS NO :1139-83-9) is an urolithin ,it is also produced in the human gut after absorption of ellagitannins from
pomegranate. Urolithin B can mitigate muscle damage experienced during intense exercise and protect muscle against stresses induced by a high-fat diet.
We incubated myotubes with different urolithin concentrations during 24 h and evaluated the impact of each treatment on their size (see Supporting Information, Figure S1A-S1D). The latter was never affected in the presence of urolithin A, whereas low doses of urolithin B increased the diameter of myotubes. The most effective concentration was identified at 15 μM as the effect disappeared with higher doses (see Supporting Information, Figure S1A and S1B). This concentration was used for the experiments reported in Figure 1,1, which confirms the anabolic properties of urolithin B in myotubes (Figure 1A1A and and1B).1B). The higher fusion index (+16.3%) indicated an enhanced fusion capability of the cells (Figure 1C).1C). The up‐regulation of the myogenic factor myogenin (P = 0.06) and the marker of differentiation, desmin14 (P < 0.05) after 5 days, confirmed the stimulation of cell differentiation in the presence of urolithin B (Figure 1D1D and and11E).
Urolithin B, gut microbiota metabolite of ellagitannins, contributes toward multiple health benefits attributed to
ellagitannin-rich foods. The influence of urolithin B on unilateral ureteral obstruction (UUO) induced renal fibrosis and TGF-β1 induced HK-2 cells and the underlying mechanism was investigated. The results showed UUO-induced kidney injury was significantly ameliorated as shown by the improvement of renal function (BUN and Cr), kidney morphology and the suppression of renal tubular
injury markers of TGF-β1, Ang II, alpha-SMA and Col IV mRNA levels. Meanwhile, the renal inflammatory induced by UUO were also strikingly attenuated in urolithin B-treated rats (TNF-α, IL-6 and MCP-1). In vitro, urolithin B treatment markedly inhibited TGF-β1-induced HK-2 cells fibrosis and proliferation, suppressed TGF-β1 and NF-κB activation. This works indicated that urolithin B contributes critically to renal inflammation in vivo and in vitro, involving the inhibition on TGF-β/Smad and TLR4/NF-кB signaling activation.
