Xanthan gum is an extracellular acidic heteropolysaccharide produced by the fermentation of xanthomonas campestris bacterium. Made from corn starch and other carbohydrates through the processes of cluturing, extracting, evaporating and grinding, it is widely used in industries such as food, oil, ming and textile.

Processing technology

 

The production technology of xanthan gum has been refined for half a century and is now relatively mature. The substrate conversion rate was 60%~ 70%, even some foreign magazines call it "benchmark product", and compare the yield of other fermentation products with it.

Xanthomonas campestris, which secretes Xanthan gum, is the pathogenic bacteria of a large number of plants such as cabbage and alfalfa Plant, straight rod-shaped, 0.4 m ~ 0.7 TZM wide, with a single flagellum, movable, gram-negative, aerobic.

The picture is the production process diagram of xanthan gum. The production of xanthan gum is affected by the composition of culture medium and culture conditions (temperature,PH value, dissolved oxygen, etc.), reactor type, operation mode (continuous or intermittent), etc.

The commonly used media are YM medium and ym-t medium. The yields of the two media are similar, but the growth curve of YM. T medium has obvious secondary growth phenomenon. The strain can grow at 25% ~ 30%.

The fermentation temperature is 28 C. Because the secreted Xanthan gum is wrapped around the cells, it hinders the transportation of nutrients and affects the growth of strains.

Therefore, in the inoculation stage, in addition to increasing the cell concentration, the yield of xanthan gum should be reduced as much as possible.

Multi step inoculation is required (the inoculation time of each step must be controlled below 7 h to avoid the formation of xanthan gum).The number of inoculation should increase with the increase of the volume of fermentation broth.The proportion of components in the fermentation broth is also an important factor affecting the yield. Carbon source (usually glucose or sucrose)

The optimum concentration is 2% ~ 4%. Too large or too small will reduce the yield of xanthan gum; The form of nitrogen source can be organic.Compounds can also be inorganic compounds. According to experience, the ideal composition ratio is sucrose (40 g / L),Citric acid (2.1 g / L), NH4NO3 (1.144 g / L), ki-i2po4 (2.866 g / L), mgc12 (0.507 g / L),Na SO4(89 mg/L),H3BO3(6 mg/L),ZnO (6mg/L),FeC13·6H2O (20 mg/L),CaCO (20 mg / L), concentrated HC1 (0.13 ml / L), and the pH value was adjusted to 7.0 by adding sodium hydroxide.

Fermentation temperature not only affects the yield of xanthan gum, but also changes the structural composition of the product. The study points out that higher temperature can increase the yield of xanthan gum, but reduce the content of pyruvate in the product. Therefore, if it is necessary to increase the yield of xanthan gum.

The temperature should be 31 ºC ~ 33%, and the temperature range should be 27 ºC ~ 31 ºC to increase the pyruvate content.

When the pH range is neutral, it is most suitable for the production of xanthan gum. With the output of the product, the acidic groups increase and the pH value increases down to about 5. The results showed that controlling the pH value in the reaction was beneficial to the growth of bacteria, but had no significant effect on the production of xanthan gum.


The type of reactor, oxygenation rate and stirring rate have corresponding empirical data, which must be determined according to specific conditions. The following data can be referred to: the stirring rate is 200 ~ 300 R / min and the air flow rate is 1 L / L · min.

In addition to the above traditional fermentation production methods, other researchers have found the amount of time required for the synthesis and assembly of xanthan gum enzymes, and cloned related genes (the combined action of 12 genes), and selected the appropriate vector, although at present,the cost of this method is high, but it is believed that through the improvement of the process, it can further reduce the cost and control the structure of the product.
