Gram staining can be used to observe the morphology of bacteria and identify bacteria;It has a certain reference value in choosing drugs.Bacteria of positive of gram orchid family name can produce external toxin, bacterium of negative of gram orchid family name can produce endotoxin, both pathogenic effect is different, also concern with pathogenic sex.
After the initial crystal violet dyeing and iodine solution mordant dyeing, the insoluble crystal violet and iodine complex was formed in the cell wall.The cell wall of Gram-positive bacteria is thicker, the layer of peptidoglycan network is more and the cross-linking is compact. During decolorization, the mesh will shrink, and it does not contain lipids, and there will be no gaps during decolorization, so the crystal violet and iodine complex can be firmly retained in the wall, so that it is still purple.The cell walls of gram-negative bacteria because of its thin, outer membrane layer, peptidoglycan layer thin high lipid content and crosslinking degree is poor, when encounter decoloring agent, mainly lipid of the outer membrane dissolved rapidly, thin and loose peptidoglycan network can't block out of crystal violet and iodine compounds, so after decolorizing shows colorless, then through red redyeing, gram-negative bacteria are red.
This product is quick and easy to dye, and the color contrast is bright.

Storage condition:
Store at room temperature, valid for 6 months.
Usage:
1.Paraffin section dewaxed to water; The bacterial smear is left to dry naturally.
2.Gran's dye A drops dyeing 10s-30s, pour away excess solution, distilled water rinse, remove float color.
3.Dry the water, drop Gram's dye B for 1min, pour the excess solution, rinse with distilled water and remove floating color.
4.Dry the water, drop Gram's dye C from the end of the slide tip to rinse the tissue for differentiation, until the Gram's dye C flowing down the section becomes colorless, then pour the excess solution and rinse with distilled water.
5.Dry the water, add Gram's dye D for about 1s, pour away the excess solution, rinse with distilled water and remove floating color.
6.Bake the slices dry in the oven (60ºC), and put the slices into three tanks of anhydrous ethanol for rapid dehydration, 1S, 3S and 5S in turn.
7.Transparent xylene for 5min, and sealed with neutral gum.Microscopic examination, image acquisition and analysis
Staining result:
Gram-positive bacteria are blue and gram-positive bacteria are red

Note:
1.The differentiation of Gram's dye C is the key step in the dyeing process, and the degree of differentiation should be paid attention to.
2.Try not to repeat staining, the differentiation solution in the strip color also changed the permeability of the cell wall, affecting the accuracy of the final results
