Pharmacological actionEssential for convertin blood sugar into energy and is involved in metabolic  actiyities in nerves. heart. and  muscles  and in the production of red blood cells.
PharmacokineticsAfter oral administration a dosa-dependert dual transpot mechanism:  Actve absorption up to concentrations of 2 μmol and passive diffusion in concentrations over 2 μmol. The elimination half-life is
approximately 4 hours. The humman body can store approximately 30 mg Thiamine.On account of the rapid  metabolisation,the reserve capacity, at 4-10 days, is very limited.
Therapeutic indicationsea tment and prevent on of Thiamine deficiency including a specific disorder called  Wernicke-korsakoff syndrome(]S)that is related tn Iow levels of  Thiamine (Thiamine defiency)and is often see in
alcoholics.
ContraindicationsHypersensitivity to the actiw substances r to ary of the excipients. Thiamie tablets  must not  be used in children and adolescents due to its hiph active substance contant.

Dosage &directions for use: One tablet once daily. In individual cases,the dose may be increased to  one tablet 3 times daily.  The tablets are to be swallowed whole with plenty of liquid after meals.

The physician in charge should decide on the duration of administration. After a maximum period of our weeks, it should be decided whether to reduce the dose.
Thiamine tablets must not be used in children and adolescents(<18 years).

Adverse reaction: Gastrointestial disorders: Unknow Castreintastral complaints such as nausea, omiting,  diarrhoea  and abdominal pain.
Immune system disorders: Vary rare: Hypersensitivity reactons such as sweating, tach cardia and skin reactions  like itching and urticaria.
Warnings& precautions for use in special populations: Large doses may interfere with the metabolism of  other B-vitamins and precipitate the symptoms of other deficiency states in poorly nourished patents

Pregnancy: During pregnancy and the nursing period the generaly recommended daily dosage of vitamin  B1 is 1.4 mg. The dosage may be exceeded in pregnant patients with  manifest Bl deficiencies only as the safety of  doses higher than the recommended daily dosage has not yet been demonstrated, The treating physician  should decide about the use of this product during pregnancy after carefully  weighing the risk-to-benefit ratio.

Nursing period: Vitamin BI is secreted into human breast milk. The advantages of breast-feeding for  the infant should be carefully weighed against the therapeutic benefit for the  women in order to decide to either discontinue breast-feeding or therapy with Thiamine.

Drug interactions: Thiamine is inactivated by 5-fluorouracil as the latter competitively inhibits the  phosphorylation of Thiamine to Thiamine pyrophosphate. Loop diuretics, e.g. furosemide that inhibit tubular
reabsorption may cause increased excretion of Thiamine in ong-term therapy and, thus, lowering of the Thiamine level. Beverages containing sulphite(e. g. wine)enhance Thiamine degradation.

Symptoms of over dosage its treatment: Thiamine has a broad therapeutic range. Very high doses  (over 10gm) have a ganglion-blocking effect, similar to that of curare, and suppress  the conduction of nerve impulses.  Symptoms of overdosage subside on withdrawal.
