Thiamphenicol is a kind of chloromycetin type broad-spectrum antibiotic,its chemical constitution is similar to the chloromycetin's,its armor sulphonebase has substituted for the chloromycetin's niryl,thus the toxicity reduces,its antibacterial function is 2.5-5 times stronger than the chloromycetin.Clinically,the Thiamphenicol mainly uses in intestinal tract surgery,department of obstetrics and gynecology and department of eye;ear;nose and throat for curing the infection,especially has good function to light and moderate infection.Thiamphenicol also has strong immunosuppression action,is one kind of fine immunosuppressant.Its immunosuppression action is much more better than chloromycetin.
Function

1.Mainly for chickens,ducks,geese spirit sluggish,enteritis,pulling yellow-white stool,airsacculitis,peritonitis,perihepatitis,salpingitis,vitelline peritonitis,the follicle deformation,intestinal mucosa congestive and bleeding,the mesentery appears granuloma accompanied with breathing difficult and other sympton caused by refractory E.coli,pasteurella anatipestifer,salmone,etc.

2.For livestock and poultrys acute or chronic respiratory disease,cough,runny nose.Also have good effect on E.coli illness,salmonella,eperythrozoonosis,staphylococci diseases,fowl cholera,necrotizing enterocolitis and other bacterial diseases.

Application

Thiamphenicol (also known as thiophenicol and dextrosulphenidol) is an antibiotic. It is the methyl-sulfonyl analogue of chloramphenicol and has a similar spectrum of activity, but is 2.5 to 5 times as potent. Like chloramphenicol, it is insoluble in water, but highly soluble in lipids. It is used in many countries as a veterinary antibiotic, but is available in China and Italy for use in humans. Its main advantage over chloramphenicol is that it has never been associated with aplastic anaemia.

Thiamphenicol is also widely used in Brazil, particularly for the treatment of sexually transmitted infections and pelvic inflammatory disease.

Unlike chloramphenicol, thiamphenicol is is not readily metabolized in cattle, poultry, sheep, or humans, but is predominantly excreted unchanged. In pigs and rats the drug is excreted both as parent drug and as thiamphenicol glucuronate.

Pharmacological Action:
Pharmacodynamics
Thiamphenicol is an amide alcohol antibacterial drug with a broad-spectrum antibacterial effect. It has a stronger effect on Gram-negative bacteria than Gram-positive bacteria. It has a stronger effect on most Enterobacteriaceae, including typhoid bacillus, paratyphoid bacillus, and Escherichia coli. Helicobacter and Salmonella are highly sensitive, and the susceptible Gram-negative bacteria include Pasteurella and Brucella. Sensitive Gram-positive bacteria include Bacillus anthracis, Streptococcus, Corynebacterium, Pneumococcus, Staphylococcus and so on. Chlamydia, Leptospira and Rickettsia are also sensitive to this product. It also has a considerable effect on anaerobic bacteria such as Clostridium tetani and actinomycetes. But Mycobacterium tuberculosis, Pseudomonas aeruginosa, and fungi are not sensitive to it.
Pharmacokinetics
This product is absorbed quickly and completely when taken orally. After being absorbed, it is widely distributed in various tissues in the body. It is mainly excreted in the urine in its original form. The half-life of cattle and sheep is shorter than that of chloramphenicol, and the half-life of pigs is 4.2 hours.

Drug Interactions:
(1) Macrolides and lincosamides have the same target of action as this product, both of which are similar to the bacterial nucleus.
Glycosome 50S subunits are combined and can produce antagonistic effects when used in combination.
(2) When combined with β-lactams, this product can produce antagonistic effects due to its rapid antibacterial effect.
(3) It has an inhibitory effect on liver microsomal drug-metabolizing enzymes, which can affect the metabolism of other drugs, increase blood drug concentration, and enhance drug efficacy or toxicity. For example, it can significantly extend the anesthesia time of pentobarbital sodium.

Indications:
Amido alcohol antibiotics. It is mainly used to treat bacterial infections such as the intestines and respiratory tracts of livestock and poultry and bacterial diseases of fish.

Dosage and Administration:
For oral administration:
Livestock and Poultry: 1-2 g for per 10 kg of body weight. Use it twice a day for 2 to 3 times.
Fish: Mixed bait feeding, 50g per 150kg body weight, once a day for 3 to 4 days.

Adverse Reactions:
(1) This product is toxic to the blood system. Although it will not cause aplastic anemia, the reversible erythropoiesis inhibition caused by it is more common than chloramphenicol.
(2) This product has a strong immunosuppressive effect, about 6 times stronger than chloramphenicol.
(3) Long-term oral administration can cause digestive disorders, symptoms of vitamin deficiency or double infection.
(4) Embryonic toxicity.
(5) It has an inhibitory effect on liver microsomal drug-metabolizing enzymes, which can affect the metabolism of other drugs, increase blood drug concentration, and enhance drug efficacy or toxicity. For example, it can significantly extend the anesthesia time of pentobarbital sodium.

Precautions:
(1) Animals with severely impaired immune function during the vaccination period are prohibited.
(2) Use with caution for livestock during pregnancy and lactation.
(3) Animals with renal insufficiency should reduce the dose or extend the interval between administrations.

Withdrawal Period:
For meat: 28 days;
For milk: 7 days;
For fish: 500 degree days.
