Olaparib is a new type of poly ADP ribose polymerase (PARP) inhibitor, including PARP1, PARP2, and PARP3. PARP mediated a DNA repair mechanism, plays an important role in DNA damage repair and apoptosis, so Ola Pani specific effect on target cell DNA damage repair mechanism, by attacking the carrying BRCA1 and BRCA2 mutation critical vulnerabilities in cancer cells play a role, can be used for breast cancer susceptibility gene (BRCA) mutation and for the maintenance treatment of platinum sensitive recurrent severe ovarian cancer patients.
In ovarian cancer and breast cancer cells, the BRCA mutation is the first hit on cell survival, because it increases the susceptibility to DNA damage. By targeting the PARP controlled backup repair pathway, Olaparib and its similar drugs have achieved a double hit on cell survival. As the two repair signaling pathways were out of balance, the accumulation of DNA damage produced a third hit on the cell.

Function & Application
Indicated for the maintenance treatment of adult patients with recurrent epithelial ovarian, fallopian tube or primary peritoneal cancer, who are in a complete or partial response to platinum-based chemotherapy.
* Indicated as monotherapy in patients with deleterious or suspected deleterious germline BRCA mutated (as detected by an FDA-approved test) advanced ovarian cancer who have been treated with three or more prior lines of chemotherapy.
* Indicated for the treatment of patients with germline breast cancer susceptibility gene (BRCA) mutated, human epidermal growth factor receptor 2 (HER2)-negative metastatic breast cancer, who have been previously treated with chemotherapy.
