N-hydroxysuccinimide, NHS, is an organic compound with the formula C4H5NO3, which is slightly acidic and irritating to skin, eyes, and mucosa.

Production method:

1. Heat 100g(1.0mol) succinic anhydride and 70g(1.0mol) hydroxylamine hydrochloride together to make the reaction system maintain negative pressure to remove the volatiles generated, rapidly heat to above 125 ° C, and slowly rise to 160 ° C after 1h.Stop heating. When the temperature drops to 125ºC, pour the liquid reactants into 400ml of ether and stir vigorously.After the product is cured, the ether layer is dumped, the cured product is coheated to boiling with 400mL butanol, and the filtrate is quickly cooled to 0ºC.After 1h of filtration, the crystals were washed successively with butanol and ethyl ether. The crude products were recrystallized with ethyl acetate, and 50g finished products were obtained with a yield of 44%.

 

Application:

N-hydroxysuccinimide and its derivatives are important raw materials for the synthesis of peptides, antibiotics, amino acids and proteins.N-hydroxysuccinimide is used to prepare active ester, inhibit the occurrence of external optical rotation when the peptide is coupled, etc. It has a very wide range of USES in medicine, and is the basic chemical raw material.Used for synthesis of amino acid protectants, semi - synthesis of kanamycin and pharmaceutical intermediates.
