Introduction:
This product is used to remove Mycoplasma cells. It is a combination reagent of quinolones and tetracyclines. It interferes with DNA replication and protein synthesis of Mycoplasma respectively, thereby effectively removing Mycoplasma cells. This product has low toxicity to eukaryotic cells and can completely eliminate the infected mycoplasma in about 10 days. After replacing the normal medium, the cell is in good condition, and there is no re-positive phenomenon in continuous multi-generation culture.

Storage and transportation
Transport with wet ice; store at -20°C to avoid repeated freezing and thawing. The validity period is 12 months.

Steps

1. Take 1 mL of Mycoplasma Elimination Reagent (100×) and add it to 100 mL of cell culture medium (containing serum, without double antibodies), mix well to obtain a medium containing Mycoplasma Elimination Reagent;

2. When mycoplasma-infected cells are passaged or changed, replace the medium containing Mycoplasma Elimination Reagent for routine cell culture;

3. After culturing for one week, inoculate a part of the cells into a 6-well plate (or other culturing utensils), and replace with normal complete medium. The remaining cells continue to be cultured with the medium containing Mycoplasma Elimination Reagent;

4. The cells in the 6-well plate are cultured for 2-3 days with normal complete medium, and when the confluence of the cells reaches 90% or more, the mycoplasma detection is performed (G1900, G1901, G1902 are recommended);

5. According to the test results, perform the following operations:

a) The result is positive: continue to repeat steps 3-4 until the test result is negative.

b) The result is negative: The cell group containing Mycoplasma Elimination Reagent medium is replaced with a normal complete medium, and mycoplasma detection is performed after a period of incubation; a negative retest result indicates that the removal is complete.

Precautions

1. This product has been tested for removal in a variety of mycoplasma-infected cell lines, but different cell lines have different sensitivity to the reagent. If the cytotoxicity is too great or the removal effect is not good, the solubility of the product can be adjusted appropriately. Other scavengers can also be used (G4007, G4008 are recommended).

2. There are many ways to infect cells with Mycoplasma. It is recommended to add mycoplasma prevention reagents in daily culture (G4008 is recommended).

3. Cells that have been determined to be mycoplasma contaminated, try to use a separate ultra-clean table during operation to avoid cross-contamination.
