Product Introduction

In histopathology, the tissue must be fixed in time and effectively before the subsequent embedding and sectioning can be completed. The role of tissue fixation is to maintain the inherent morphology and structure of tissues and cells, prevent bacterial corrosion and autolysis of tissue cells, preserve the inherent substances in cells, coagulate intracellular proteins, and minimize or terminate endogenous and exogenous The enzyme reaction keeps the cells or tissues basically the same as the substances in life, and can also harden the tissues and facilitate the cutting of the tissues. For some infectious specimens, it can prevent the spread of diseases; keep the general specimens. Finally, fixation can also enhance the effect of dyeing. Glutaraldehyde is a commonly used fixative, which preserves the fine structure of cells well, but has weak penetrating ability. It is mainly used for the early fixation of electron microscope samples.

This product is a fixing solution for electron microscopy, the active ingredient is 2.5% glutaraldehyde, the solvent is 0.1 M phosphate buffer, and the pH is 7.0-7.5@25°C.

Storage and transportation

Transported in wet ice; stored at 4°C and protected from light, valid for 24 months.

Instructions for use

The fresh tissue was quickly cut into 1-2 mm3 size, immersed in a sufficient amount of this fixative, and fixed at 4°C for 2-4 h. If the tissue contains a lot of blood and tissue fluid, the tissue should be washed several times with fixative, and then quickly cut into small pieces for fixation. If it is not possible to cut into 1-2 mm3 tissue blocks at one time, you can also cut into slightly larger tissue blocks, and then fix them into 1-2 mm3 size.

Precautions

1. Take the material quickly, and ensure that the tissue is placed in the pre-cooled electron microscope fixative within 1 min after being removed from the living body. The operation is best performed at low temperature (0-4°C) to reduce enzyme activity and prevent cell autolysis.

2. The tissue volume should be as small as possible, not more than 2 mm3, to ensure that the fixative is fully penetrated into the tissue cells to obtain a good fixation effect. When trimming the tissue block, try to do it in the fixative.

3. Use sharp dissecting instruments when taking materials to avoid damage to the ultrastructure caused by tissue traction, contusion and extrusion.

4. Please use it in a well-ventilated environment, and protect it from inhalation.

5. Please tighten the cap in time after use to prevent the active ingredients from volatilizing.

6. For your safety and health, please wear lab coats and disposable gloves for operation.
