Product Introduction
The AB-PAS dye solution of this product is a combination of Alcian Blue (AB) and PAS dye solution. Among them, Alcian blue, also known as Alcian blue, is a kind of copper-like peptide cyanine dye, which is alkaline and belongs to cationic dye. The dye can combine with acidic groups to color tissues with anionic groups (such as carboxyl groups). Alcian Blue is a specific dye that displays acidic mucus, which makes the acid mucopolysaccharide in the cytoplasm appear blue, while the neutral mucin is not colored. PAS staining solution is used for glycogen PAS staining (Periodc Acid-Schiff stain), also known as perioic acid-Schiff stain, which is mainly used to detect glycogen and polysaccharides in tissues in tissue morphology. The main principle is to use the oxidant perodic acid to oxidize the hydroxyl groups on the adjacent two carbon atoms in the carbohydrates to aldehyde groups, and then use Schiff reagent to react with the aldehyde groups to form a purple-red substance. Alcian Blue and PAS staining solution combined staining can distinguish acid glycoproteins and neutral glycoproteins.
This product is a set combination. AB-PAS dye solution A is Schiff Schiff reagent, AB-PAS dye solution B is 0.5% GDS solution, and AB-PAS dye solution C is alcian blue with a concentration of 1% and a pH of about 2.5. After the sample is dyed with this product, the acidic mucous substance is blue, glycogen and neutral mucous substances are red, and the mixed mucous substance is blue-purple or purple-blue. Generally speaking, in normal gastrointestinal mucus, the surface epithelium of gastric mucosa, pyloric glands, duodenal glands, etc. are mainly red neutral mucus substances. The goblet cells and intestinal glands of the small and large intestine mucosa mainly secrete acidic mucus.
Storage and transportation
Transport in wet ice; the whole set of reagents is stored at 4°C, and the validity period is 12 months. Among them, AB-PAS dye solution B and AB-PAS dye solution C can also be stored at room temperature.
Preparation before experiment
Bring your own xylene, gradient ethanol, absolute ethanol, neutral gum, etc.
Instructions
1. Paraffin sections are deparaffinized to water: sections are sequentially deparaffinized with xylene for 10 minutes, replaced with fresh xylene for 10 minutes, absolute ethanol for 5 minutes, fresh absolute ethanol for 5 minutes, 90% ethanol for 5 minutes, and 75% ethanol 5 min, wash with tap water.
2. Dip the section into AB-PAS dye solution C for 8-10 min, and wash with tap water until the slide is colorless.
3. The slices are immersed in AB-PAS dye solution B for 10-15 min, and then immersed in pure water for 3 times, each time about 10 s.
4. The sections are placed in AB-PAS dye solution A (return to room temperature in advance), covered and protected from light for 25-30 min, and rinsed with running water for 5 min.
5. Cut slices into three cylinders of absolute ethanol for dehydration, each time for 5 minutes; pass xylene to clear for 5 minutes, replace with fresh xylene and then clear for 5 minutes, then cover with neutral gum.
Precautions
1. AB-PAS dye solution B can be reused. The dye solution is transparent. If it is obviously yellow, has impurities, or the staining of the tissue is too light, it needs to be replaced with a new dye solution.
2. AB-PAS dye solution A needs to be stored at 4°C, take it out and return to room temperature before use. When the color of the solution is obviously red, it is recommended to replace with a new dye solution.
3. Please wear lab coat and disposable gloves during operation.
