Daptomycin is a lipopeptide antibiotic produced by Streptomyces roseosporus with reported in vitro bactericidal activity against gram-positive organisms. As a calcium-dependent antibiotic, Daptomycin has been reported to act on the cytoplasmic membrane and undergo a conformational change upon interaction with lipid membranes in a model system. After the conformational change occurs, the compound is inserted into the membrane and causes significant perturbations. Membrane depolarization and leakage occurs, causing death in susceptible organisms.

Daptomycin is from Streptomyces (S.reseosporus) extracted from cyclic lipopeptide antibiotics a new structure of the fermentation broth, it not only has a novel chemical structure, and the mode of action and any one of the approved antibiotics are different: it is through the transport of amino acids disrupt the cell membrane, thus impeding the biosynthesis of bacterial cell wall peptidoglycan, change the nature of cell membrane, can damage the function of bacterial cell membrane in many aspects, and quickly kill gram positive bacteria.
In addition to the role of daptomycin in most clinically relevant gram positive bacteria, it is more important to have in vitro showed methicillin resistant (methicillin) strains, vancomycin and linezolid resistant properties with potent activity for this feature has very important clinical significance in patients with severe infection.
The research shows that the antibacterial spectrum of activity is similar to vancomycin and daptomycin, mainly has a strong antimicrobial effect on Gram positive bacteria, such as Staphylococcus aureus spp MIC=0.125 ~ 0.5 g/ml, the gram bacteria spp MIC=0.06 ~ 0.5 g/ml, 0.25 MIC= of Enterococcus to 2 mu g/ ml. Daptomycin has broad-spectrum antibacterial activity in the gram positive anaerobic bacteria on spp. MIC=0.12, Peptostreptococcus g/ml, spp.MIC=0.5 of Clostridium g/ml of Lactobacillus spp.MIC=1 g/ml.
