WHAT IS sm04554?

sm04554 is a molecule that acts at the level of the skin androgen receptor, blocking dihydrotestosterone (DHT) from binding to the receptor in the cell. The molecule has a unique mechanism that acts at the skin surface, and is quickly metabolized to free inactive cortexolone, a substance naturally found in the body. The molecule has been evaluated in a variety of animal models as well as initial human trials in Europe for acne and androgenic alopecia. The results of these studies demonstrate that sm04554 appears to have potent anti-androgenic activity without significant local or systemic side effects.

Function

SM04554 reduces eosinophilic airway inflammation and is well tolerated in patients with persistent moderate-to-severe asthma and raised sputum eosinophil counts despite inhaled corticosteroid treatment. SM04554 demonstrates a favorable safety profile.

Application

SM04554 is an oral active and potent CRTh2 receptor antagonist and potentially useful for asthma treatment.
SM04554 displayed high affinity for the human CRTh2 receptor (1.14 ± 0.44 nM) expressed in Chinese hamster ovary cells, the binding being reversible and competitive with the native agonist prostaglandin D2(PGD2). SM 04554 was also a potent inhibitor of PGD2-induced cytokine release in human Th2 cells.
