Recombinant HRV 3C Protease (Human Rhinovirus 3C Protease,Prescission, Human Rhinovirus type 14 3C Protease, a commonly used cysteine Protease, is a recombinant expression of His-tagged Human Rhinovirus type 14 3C Protease in Escherichia coli.It can specifically recognize the sequence L-E-V-L-F-Q-G-P (Leu-Glu-Val-Leu-Phe-Gly-Gly-Pro) or the core pentapeptide sequence L-F-Q-G-P (Leu-Phe-Gly-Gly-Pro) at low temperature.The enzyme digestion was performed between Q (Gln) and G-P (Gly-Pro) amino acid residues.The enzyme had strong specificity and high activity, and it still had high activity even at 4ºC.
Application: It is often used to remove MBP, GST, HIS or other tagged proteins from fusion proteins.The protease itself carries the HIS label and binds to the nickel column.

Definition of enzyme activity: 100 μg of substrate (protein containing HRV 3C Protease digestion site) can be cut more than 90% of the required enzyme activity unit within 16h under the condition of 4ºC, pH 7.5.
Inactivation or inhibition: 100mM ZnCl2, 4mM AEBSF and 100μm Chymostatin all inhibit the activity of HRV 3C Protease.
Purity and concentration: SDS-PAGE purity ≥95%, 2 U/μL.
Recombinant HRV 3C Protease Storage solution:50 mM Tris-HCl,150 mM NaCl,1 mM EDTA,1 mM DTT,50% (v/v) glycerol,pH 8.0.
10×HRV 3C Buffer:500 mM Tris-HCl,1.5 M NaCl,10 mM EDTA,10 mM DTT,pH 7.5.
Storage and transportation:
Transport with wet ice; store at -20ºC, valid for 12 months.

Operating steps
1. Protenase digestion system recommendation:

2. The reaction system was placed at 4ºC for overnight reaction or 16h;
3. 10μL sample of digestion product was taken for SDS-PAGE detection to evaluate the amount of digestion and reaction conditions.

Notes:
1. Enzyme products should be placed in an ice box or ice bath, and stored at -20ºC immediately after use. Separate storage is recommended.
2. For your safety and health, please wear a lab coat and disposable gloves to operate.
