Pharmacology and Toxicology:
Pharmacology: This product is a derivative of artemisinin, which has a strong and rapid killing effect on the erythrocyte stage of Plasmodium, and can quickly control clinical attacks and symptoms. The mechanism of action of artemisinin is still unclear, mainly interfering with the function of surface membrane mitochondria of Plasmodium. Artemisinin alters the membrane structure by affecting the ultrastructure of the erythrocyte phase of Plasmodium. Because of the role of food vesicles, the nutritional uptake of Plasmodium is blocked. When Plasmodium loses a large amount of cytoplasm and nutrients, but can not be supplemented, it quickly dies. Its action is mediated by free iron produced by the decomposition of hemoglobin through its peroxide (hydrogen peroxide) bridge, which produces unstable organic free radicals and/or other electrophilic intermediates, and then forms covalent adducts with the proteins of Plasmodium, resulting in the death of Plasmodium. The antimalarial activity of artemether was 6 times higher than that of artemisinin. Toxicology: Studies on animal reproductive toxicity have shown that administration during pregnancy induction period in mice can increase embryo absorption, but has no teratogenic effect.
Pharmacokinetics:
Oral absorption is good and effective. After oral administration of 2 mg/kg dihydroartemisinin, the peak blood concentration reached 1.33 hours, and the maximum blood concentration was 0.71 mg/L. The plasma T1/2 was 1.57 hours. It is widely distributed in the body and excretes and metabolizes rapidly.
