Principle
Amyloid proteins are easily stained by Congo red due to the selective affinity. Congo red is a linear diazo dye, binds the linear arranged amyloid proteins or fibrils through hydrogen bonds. When viewed with polarized light microscopy, red to green birefringence occurs due to the parallel alignment of the dye molecular on these linearly arranged amyloid proteins or fibrils.

Preservation
Valid in 3 months at room temperature.
Procedure
1,deparaffinize and rehydrate to distilled water. 
2,stain in Congo red solution for 1 h, wash in running tap water 2 min. 
3,differentiate in alkaline alcohol solution until positive plaque occurs in colorless background, then wash in tap water.
4,rinse in hematoxylin solution for 1-3 min, differentiate in hydrocloric acid solution for a few seconds, wash in tap water and then back to blue in amonia solution. Wash in running tap water.
5,dehydrate 3 changes in absolute alcohol, 5 min each. Clear 2 changes in xylene, 5 min each. Then mount with resin mounting medium.
6,Microscopy determination and analysis.
Amyloid proteins and elastic fibers are red. Nucleus are light blue.


Notes

1,when stain amyloid protein using Congo red, thyrocolloid and elastic fibers are also easily colorized in red and sometimes display collagen fibers in red. Be sure to distinguish them according to the morphological structure.
2,manage differentiate in alkaline alcohol in a proper degree by observing in microscopy. Inadequate differentiate result in collagen fibers to be stained with red. While over-differentiate may result in amyloid proteins decolorization. If over-differentiate happen, re-stain slides from step 2.
