Product Introduction

Amyloid is an extracellular eosinophilic substance with no fixed shape. It is mainly composed of protein. Most of the protein is arranged in a reverse β-sheet structure. Such amyloid can exist in different tissues and organs. The resulting disease is called amyloidosis. Congo red can be used for the color development of amyloid in living organisms and tissue slices. The basic principle is that amyloid has a high affinity for Congo red dye, and its hydroxyl group binds to the amino group of Congo red to dye the amyloid red. Congo red staining is a classic amyloid staining method.

This product is composed of Congo red dye and alkaline differentiation solution, which can be used for staining of amyloid. After staining, the amyloid plaques are light pink to orange-red clouds or lumps, and the amyloids also show a yellow-green birefringence under polarized light.
Instructions for use

If you need to counter-stain cell nuclei, prepare your own hematoxylin staining solution, hematoxylin differentiation solution and hematoxylin returning blue solution. Bring your own ethanol, xylene, neutral gum, etc.
Congo Red Dye Solution for Staining of Amyloid 250ml
1. Paraffin sections are deparaffinized and rehydrated according to conventional procedures.

2. Congo red staining: the sections are soaked in Congo red staining solution overnight, and then washed with tap water for 2 min.

3. Differentiation: The slices are differentiated into positive amyloid plaques by alkaline differentiation solution, the background is basically colorless, and then washed with tap water.

4. (Optional) Staining nuclei: Stain the slices in hematoxylin staining solution (G1004) for 1 min, differentiate with hematoxylin differentiation solution (G1039), wash with tap water, return to blue with hematoxylin blue solution (G1040), and rinse with running water.

5. Dehydration and mounting: the slices are dehydrated three times with absolute ethanol for 5 minutes each time; the sections are transparent with xylene twice, each time for 5 minutes. Add neutral gum to seal the sheet.
