Endo Cave hydrate is a new type of cyclopentylguanosine anti HBV drug. Its pharmacological action is similar to that of entredo Cave. It is clinically suitable for the treatment of chronic hepatitis B virus inflammation with active replication of the virus, continuous elevation of serum aminotransferase ALT or liver histology showing active lesions. This product is guanine nucleoside analogue and has inhibitory effect on hepatitis B virus (HBV) polymerase. It can be phosphorylated into active triphosphate. The half-life of triphosphate in cells is 15 hours. By competing with deoxyguanine nucleoside triphosphate, the natural substrate of HBV polymerase, entecavir triphosphate can inhibit all three activities of viral polymerase (reverse transcriptase): (1) the initiation of HBV polymerase. (2) Pre genomic mRNA reverse transcription negative chain formation. (3) Synthesis of HBV DNA positive chain. The inhibition constant (KI) of entecavir triphosphate on HBV DNA polymerase was 0.0012 μ m. Effects of entecavir triphosphate on cells α,β,δ DNA polymerase and mitochondria γ The inhibition of DNA polymerase is weak, and the Ki value is 18 to 160 μ m.
